Simultaneous LC-MS/MS quantitation of semaglutide and cagrilintide enables pharmacokinetic evaluation of a novel nanoparticle oral co-delivery platform

Scientific poster | Presented at AAPS PharmSci 360 2026

Developing oral combination peptide therapeutics requires innovative formulation technologies supported by sensitive, reliable bioanalytical methods. For compounds such as semaglutide and cagrilintide, the ability to measure both peptides simultaneously is essential for evaluating their pharmacokinetic behavior following co-administration.

In research presented at AAPS PharmSci 360 2026, Veloxity Labs and Toralgen demonstrated how simultaneous LC-MS/MS quantitation supported preclinical evaluation of a novel nanoparticle-based oral co-delivery platform.

Toralgen’s formulation incorporated semaglutide and cagrilintide into separate populations of polymerized ursodeoxycholic acid (pUDCA) nanoparticles, combined into a single oral tablet. Veloxity developed an LC-MS/MS method capable of quantifying both peptides in beagle dog plasma across a 2.00–1,000 ng/mL calibration range.

Following oral administration of the formulation, both peptides were measurable in plasma, providing initial evidence of systemic exposure and supporting further investigation of the co-delivery platform.
