How simultaneous LC-MS/MS quantitation supported preclinical evaluation of semaglutide and cagrilintide in a novel nanoparticle-based oral delivery platform
Oral peptide therapeutics hold significant promise for expanding treatment options, particularly in therapeutic areas such as obesity and metabolic disease. However, challenges associated with peptide stability, absorption and bioavailability continue to drive the need for innovative delivery technologies and specialized bioanalytical approaches.
In a collaborative research effort presented at AAPS PharmSci 360 2026, Veloxity Labs and Toralgen investigated a novel approach to oral co-delivery of semaglutide, a GLP-1 receptor agonist, and cagrilintide, an amylin analog. Toralgen’s nanoparticle-based formulation brought both therapeutic peptides together in a single oral tablet, while Veloxity’s simultaneous LC-MS/MS method enabled measurement of both compounds from the same plasma sample. The resulting preclinical findings demonstrated measurable systemic exposure to both peptides following oral administration in beagle dogs, providing initial proof-of-concept evidence for the formulation platform.
